So you've been able to print in the past from LR4 and CS6 and this problem has just started?
Made any hardware or software changes that may have precipitated the problem?
Have you tried using a different cable to the printer?
Still sounds like ink or printer head to me too. I don't have any experience with third party inks though. How much ink is in the magenta cartridges? Can you change them?
I agree with John, you still end up with a tiff file, but not only can you edit the Nik adjustments, you can still make changes to the raw file in ACR.
Does sound like an ink problem. If you've checked all the print settings and they seem ok, try printing from a different application and see if that makes a difference.
I agree with Mark's advice. I'd definitely check to confirm the files are what you think then backup anything you're deleting elsewhere. You can get rid of the backups once you're satisfied you've got everything the way you want it.

When I change colour management to "Managed by Printer" I do get access to the colour matching dialogue box, provided I don't have a preset set in the print dialogue. That's tripped me up in the past which was why I suggested it.

There are a couple of benefits to writing to .xmp. If for some reason you lose your whole catalog, you will still have the changes you have made. Additionally, if you're backing up incrementally, only the .xmp files need to be backed up after changes are made, not the image files.
Welcome to the forum. I was in shakeytown last weekend. Horrible to see it the way it is now, but at least things seem to have settled down a bit.
LR4 is great, a huge improvement on v3, I'm sure you'll love it.
In the library module, you should be able to see the folder locations on the left hand toolbar.
When you import the files, Lightroom gives you the option of moving, copying, or leaving the files in their original location.
What paper size do you have selected? The image above looks like a 4x6 on A4 or similar sized paper. Select 4x6 paper from the Page Setup and you should see a better result. To print a landscape orientation image, select "Rotate to Fit" from the image settings on the right hand toolbar.
